Gene Infomation:Bcl-6, a transcriptional repressor, binds Stat recognition-like DNA elements and influences germinal center development and cell differentiation. Additionally, Bcl-6 negatively regulates NFκB expression, thereby inhibiting NFκB-mediated cellular functions.HDAC- and silent information regulator (SIR)-2-dependent acet-ylation of Bcl-6 causes downregulation of activity by inhibiting the ability of Bcl-6 to recruit complexes containing histone deacetylases (HDACs). Bcl-6 is frequently deregulated in non-Hodgkin’s B cell lymphomas. Bcl-6b (B-cell CLL/lymphoma 6, member B), also known as ZNF62, BAZF or ZBTB28, is a 480 amino acid nuclear protein that contains one BTB (POZ) domain and five l C2H2---type zinc fingers. Expressed ubiquitously with highest expression in placenta and heart, Bcl-6b associates with Bcl-6 and functions as a sequence-specific transcriptional repressor that is thought to be necessary for early B-cell development.

Background:The NF-kappa-B (NFKB) complex of proteins is inhibited by I-kappa-B (IKB) proteins, which inactivate NFKB by trapping it in the cytoplasm. Phosphorylation of serine residues on the IKB proteins by IKB kinases marks them for destruction via the ubiquitination pathway, thereby allowing activation and nuclear translocation of the NFKB complex.
