Background:1. 取100 μl EDTA抗凝的人外周血加入5 ml流式管底;2. 取10 μl标记抗体加入流式管底与全血混匀,室温避光孵育20分钟;3. 加入2ml 1×红细胞裂解液混匀后避光放置10分钟,溶解红细胞;4. 将样品管置离心机中以1000 rpm离心5分钟后弃上清;5. 加入2 ml PBS洗液重悬细胞,1000 rpm离心5分钟,弃上清;6. 加入SAV-PE或SAV-FITC,室温避光孵育20分钟;7. 加入2 ml PBS洗液重悬细胞,1000 rpm离心5分钟,弃上清;8. 加入0.5 ml PBS洗液重悬细胞,待流式细胞仪检测(样品应在当天上机测定,也可加固定液4℃保存过夜测定).[PBS洗液:PBS+1%FBS+0.1%NaN3][细胞固定液:2%甲醛溶液]
Molar Function:识别一个140 kDa 的特异性B淋巴细胞抗原CD22。CD22抗原存在于不同阶段发育的B淋巴细胞胞浆内,在成熟B淋巴细胞,仅表达于细胞表面。CD22抗原也表达在大多数B细胞白血病和淋巴瘤;但在任何T细胞白血病不表达。CD22分子参与B细胞激活作用。与其它B淋巴细胞抗原如CD10,CD19和CD20不同,CD22抗原还存在于浆细胞样淋巴细胞上,但在完全成熟的浆细胞上减少。CD22是黏附分子,在B细胞活化过程中作为信号分子而发挥重要作用。CD22抗体可用于治疗白血病和淋巴瘤。
Western Blot:Stability: The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37°C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.
Storage: Store at 4°C for frequent use. Stored at -20°C in a manual defrost freezer for one year without detectable loss of activity. Avoid repeated freeze-thaw cycles.
