Background:1. 取100μl EDTA抗凝的人外周血加入流式测定管底部;2. 加入适量纯化抗体与全血混匀,室温孵育30分钟;3. 加入2 ml 1×红细胞裂解液混匀后避光置10分钟,溶解红细胞;4. 将试管置离心机中以1000 rpm离心5分钟后弃上清;5. 加入2 ml PBS洗液重悬细胞以1000 rpm离心5分钟,弃上清;6. 加入适量荧光标记抗小鼠免疫球蛋白试剂,室温避光孵育20分钟;7. 加入2 ml PBS洗液重悬细胞以1000 rpm离心5分钟,弃上清;8. 加入0.5 ml PBS洗液重悬细胞,待流式细胞仪检测(样品应在当天上机测定,也可加固定液4℃保存过夜测定).[PBS洗液:PBS+1%FBS+0.1%NaN3][细胞固定液:2%甲醛溶液]

Molar Function:3E1-D1单抗识别CD33抗原,一个67 kDa的I型跨膜糖蛋白,表达于髓系前体细胞、单核细胞和粒细胞表面,淋巴细胞、血小板、红细胞、造血干细胞和非造血系统不表达。CD33抗原是依赖硅铝酸的细胞黏附分子,参与人自我更新的造血干细胞的阴性选择。此克隆与非人灵长类动物有交叉反应。可用于急性髓细胞白血病的免疫分型。

Western Blot: 

Stability: The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37°C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.

Storage: Store at 4°C for frequent use. Stored at -20°C in a manual defrost freezer for one year without detectable loss of activity. Avoid repeated freeze-thaw cycles.
