Background:1. 取100 μl EDTA抗凝的人外周血加入5 ml流式管底;2. 取10 μl标记抗体加入流式管底与全血混匀,室温避光孵育20分钟;3. 加入2ml 1×红细胞裂解液混匀后避光放置10分钟,溶解红细胞;4. 将样品管置离心机中以1000 rpm离心5分钟后弃上清;5. 加入2 ml PBS洗液重悬细胞,1000 rpm离心5分钟,弃上清;6. 加入0.5 ml PBS洗液重悬细胞,待流式细胞仪检测(样品应在当天上机测定,也可加固定液4℃保存过夜测定).[PBS洗液:PBS+1%FBS+0.1%NaN3][细胞固定液:2%甲醛溶液]

Molar Function:1D12-H8单抗识别CD38抗原,一个45 kDa膜整合糖蛋白,广泛表达于所有前B细胞、浆细胞、胸腺细胞、激活的T细胞、NK细胞、单核/巨嗜细胞和树突细胞上。CD38抗原也存在于90% CD34+细胞,但造血干细胞不表达。CD38 + 和CD34+的共同表达说明这些细胞具有系特异性。CD38还可作为胞外酶催化多种反应,在调节细胞激活和依赖细胞环境的增殖中发挥重要作用。应用:白血病免疫分型和分类;监测HIV-1感染及疾病进展。

Western Blot: 

Stability: The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37°C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.

Storage: Store at 4°C for frequent use. Stored at -20°C in a manual defrost freezer for one year without detectable loss of activity. Avoid repeated freeze-thaw cycles.
