Background:1. 取100μl EDTA抗凝的人外周血加入流式测定管底部;2. 加入适量纯化抗体与全血混匀，室温孵育30分钟;3. 加入2 ml 1×红细胞裂解液混匀后避光置10分钟,溶解红细胞;4. 将试管置离心机中以1000 rpm离心5分钟后弃上清;5. 加入2 ml PBS洗液重悬细胞以1000 rpm离心5分钟,弃上清;6. 加入适量荧光标记抗小鼠免疫球蛋白试剂，室温避光孵育20分钟;7. 加入2 ml PBS洗液重悬细胞以1000 rpm离心5分钟,弃上清;8. 加入0.5 ml PBS洗液重悬细胞,待流式细胞仪检测(样品应在当天上机测定,也可加固定液4℃保存过夜测定).[PBS洗液:PBS+1%FBS+0.1%NaN3][细胞固定液:2%甲醛溶液]

Molar Function:7G2-C10单抗识别CD40( Bp50 )分子，TNF受体家族的一个成员，分子量为48 kDa。CD40是全B细胞标记，表达于从前B细胞到浆细胞各个B细胞发育阶段、树突细胞和CD34+造血祖细胞，但单核细胞和粒细胞不表达。CD40分子参与B细胞的生长、分化；Ig的异型转换；是树突细胞的上调黏附分子；促进巨噬细胞和树突细胞分泌细胞因子。据报道，CD40抗体可与抗-μ或佛波醇酯共同刺激B细胞增殖；用于监测移植排斥及T细胞和Ab介导的自身免疫病。此克隆与非人灵长类动物有交叉反应。

Western Blot:

Stability: The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37°C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.

Storage: Store at 4°C for frequent use. Stored at -20°C in a manual defrost freezer for one year without detectable loss of activity. Avoid repeated freeze-thaw cycles.
