Background:1. 分离血小板:将全血样本以800 rpm离心15分钟,取上清以2000 rpm离心5分钟,弃上清;2. 用PBS洗液调节血小板浓度约1×106个/ml备用;3. 准确取100ul调节好浓度的血小板悬液到流式测定管底部;4. 加入适量纯化抗体(需滴定)与全血混匀,室温孵育30分钟;5. 向样本管中加入2ml PBS洗液,混匀后以1000rpm离心5分钟,弃上清;6、加入适量荧光标记抗小鼠免疫球蛋白试剂,室温避光孵育20分钟;6. 加入2 ml PBS洗液重悬细胞,1000 rpm离心5分钟,弃上清;7. 加入0.5 ml PBS洗液重悬细胞,待流式细胞仪检测(样品应在当天上机测定,也可加固定液4℃保存过夜测定).[PBS洗液:PBS+1%FBS+0.1%NaN3][细胞固定液:2%甲醛溶液]
Molar Function:4H7-B6单抗识别钙依赖性CD41/CD61(GPⅡb/Ⅲa)复合物,分子量为135/120 kDa,表达于正常血小板和巨核细胞上。CD41抗原是纤维蛋白原、vWf、纤维蛋白酶原和玻璃体结合蛋白的受体,并介导血小板黏附和聚集。4H7-B6单抗可完全抑制ADP、肾上腺素和胶原蛋白介导的血小板激活,并部分抑制软脂酸十六酯和凝血酶诱导的血小板激活,在血小板和巨核细胞的形态学和生理学研究中非常有用。CD41可用于Glanzmann's血小板机能不全和巨核细胞白血病的诊断,同种抗体缺失及抗血栓治疗。
Western Blot:Stability: The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37°C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.
Storage: Store at 4°C for frequent use. Stored at -20°C in a manual defrost freezer for one year without detectable loss of activity. Avoid repeated freeze-thaw cycles.
