Background:1. 取100 μl EDTA抗凝的人外周血加入5 ml流式管底;2. 取10 μl标记抗体加入流式管底与全血混匀,室温避光孵育20分钟;3. 加入2ml 1×红细胞裂解液混匀后避光放置10分钟,溶解红细胞;4. 将样品管置离心机中以1000 rpm离心5分钟后弃上清;5. 加入2 ml PBS洗液重悬细胞,1000 rpm离心5分钟,弃上清;6. 加入0.5 ml PBS洗液重悬细胞,待流式细胞仪检测(样品应在当天上机测定,也可加固定液4℃保存过夜测定).[PBS洗液:PBS+1%FBS+0.1%NaN3][细胞固定液:2%甲醛溶液]
Molar Function:1C5-F11单克隆抗体识别58 kDa的T细胞抗原分子,CD5抗原是清道夫受体超家族的一个成员,在胸腺细胞和所有成熟T淋巴细胞上表达,也存在于一小部分在婴儿期增殖的成熟B淋巴细胞(B1a 细胞)和一些自身免疫性疾病和某些慢性B淋巴细胞白血病(B-CLL)细胞上。CD5分子为一双重受体,既是胸腺细胞和B1a细胞的抑制性信号受体,也是共刺激信号受体。CD5介导的细胞间反应可能影响胸腺细胞的成熟和选择。CD5也是一些B淋巴细胞增殖性疾病(B-CLL、外周区淋巴瘤、毛细胞白血病等)的表型标记。外周血CD3 +/ CD5-T细胞增加与GVHD的出现相一致。
Western Blot:Stability: The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37°C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.
Storage: Store at 4°C for frequent use. Stored at -20°C in a manual defrost freezer for one year without detectable loss of activity. Avoid repeated freeze-thaw cycles.
