Background:1. 取100 μl EDTA抗凝的人外周血加入5 ml流式管底;2. 取10 μl标记抗体加入流式管底与全血混匀,室温避光孵育20分钟;3. 加入2ml 1×红细胞裂解液混匀后避光放置10分钟,溶解红细胞(推荐: RBC Lysing Solution[10×]);4. 将样品管置离心机中以1000 rpm离心5分钟后弃上清;5. 加入2 ml PBS洗液重悬细胞，1000 rpm离心5分钟,弃上清;6. 加入0.5 ml PBS洗液重悬细胞,待流式细胞仪检测(样品应在当天上机测定,也可加固定液4℃保存过夜测定).[PBS洗液:PBS+1%FBS+0.1%NaN3][细胞固定液:2%甲醛溶液]

Molar Function:别名为B7、B7.1或BB1，分子量为60kD,表达于活化B淋巴细胞、活化T淋巴细胞、巨噬细胞、外周血单核细胞及树突状细胞。他属于免疫球蛋白超家族，其配体是CD28和CD152（CTLA4）。CD80是CD86活化T淋巴细胞时的协同刺激因子，在自身免疫监控、体液免疫应答及移植反应中发挥重要作用。CD80是T淋巴细胞活化时必需的膜抗原。T淋巴细胞活化时，T淋巴细胞的TCR/CD3和APC的MHC结合。CD80在活化B淋巴细胞、活化T淋巴细胞、人类T淋巴细胞白血病病毒-1(human T cell leukemia virus-1,HTLV-1)阳性T淋巴细胞、受IFN-γ刺激的单核细胞、树突状细胞上均有表达，而非活化B淋巴细胞、红细胞、粒细胞及核细胞上不表达。

Western Blot:

Stability: The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37°C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.

Storage: Store at 4°C for frequent use. Stored at -20°C in a manual defrost freezer for one year without detectable loss of activity. Avoid repeated freeze-thaw cycles.
