Background:1. 取100μl EDTA抗凝的人外周血加入流式测定管底部;2. 加入2 ml 1×红细胞裂解液混匀后避光置10分钟,溶解红细胞(推荐: RBC Lysing Solution[10×]);3. 将试管置离心机中以1000 rpm离心5分钟后弃上清;4. 加入2 ml PBS洗液重悬细胞以1000 rpm离心5分钟,弃上清;5. 加入1ml 固定液，吹打均匀，固定40min；6. 加入1ml 固定破膜剂，吹打均匀，1000 rpm离心5分钟,弃上清;7. 加入1ml 固定破膜剂，吹打均匀，室温孵育30min；8. 加入 1ml固定破膜剂，1000 rpm离心5分钟,弃上清;9. 加入适量抗体与全血混匀，室温孵育30分钟;10. 加入 1ml固定破膜剂，1000 rpm离心5分钟,弃上清;;11. 加入2 ml PBS洗液重悬细胞以1000 rpm离心5分钟,弃上清;12. 加入0.5 ml PBS洗液重悬细胞,待流式细胞仪检测(样品应在当天上机测定,也可加固定液4℃保存过夜测定).[PBS洗液:PBS+1%FBS+0.1%NaN3][细胞固定液:2%甲醛溶液]

Molar Function:特异性识别髓过氧化物酶 (MPO), 存在于骨髓细胞嗜天青颗粒中的一种过氧化物酶。MPO是一种多亚基蛋白，包含两个55KD亚基和两个15KD亚基。较大的亚基与血红素蛋白结合在一起，导致呈绿色荧光。作为酶制剂，MPO 分解过氧化氢和氧化酪氨酸，产生次氯酸和酪胺酰，导致中性粒细胞细胞毒性和杀伤效应。此外，髓过氧化物酶在一些癌症诊断中非常重要。

Western Blot:

Stability: The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37°C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.

Storage: Store at 4°C for frequent use. Stored at -20°C in a manual defrost freezer for one year without detectable loss of activity. Avoid repeated freeze-thaw cycles.
