Background:1. 使用淋巴细胞分离液从抗凝血中提取外周血单个核细胞 (PBMC)。PBMC用PBS, 5%BSA*, 0.09%NaN3, pH7.4洗3次,40-70µm尼龙网过滤去除细胞团块和碎片后调整细胞数为1-3uf0b4107/ml。 2. 将50µl抗人CD4抗体免疫磁珠与1uf0b4107 PBMC混合,室温反应30min, 间隔10min轻轻晃动反应管,避免磁珠沉淀,使磁珠与细胞充分反应,尽量防止出现气泡。 3. 磁分离器分离3-5min,上清为去除了CD4+T细胞和单核细胞的阴性部分。4. 1ml磁珠分选缓冲液重悬结合了CD4+细胞的免疫磁珠,玻璃滴管或移液枪枪头反复吹打磁珠,磁分离器分离3-5min,弃上清, 重复清洗4-5次。5. 清洗后的免疫磁珠可根据具体实验需要重悬于适宜的缓冲液或培养基中, 可直接用于流式检测,或者37oC培养48hrs可使细胞与磁珠分离。
Molar Function:CD4分子是单链跨膜糖蛋白,分子量为59kDa,主要表达于外周血辅助性T细胞和全部单核细胞。但CD4分子在单核细胞表面的密度明显低于辅助性T 细胞。CD4分子在胞膜外有4个结构区,其中第一结构区是人类免疫缺陷病毒(HIV)外壳蛋白gp120 识别的部位,因此CD4分子也是HIV的受体。CD4分子还可与抗原提呈细胞 (APC)表面的MHCⅡ类抗原非多态部分相结合,协助辅助性T细胞细胞识别APC细胞表面外来抗原与MHCⅡ类抗原的复合物。抗人CD4抗体免疫磁珠可从混合细胞中阳性分选或去除MHC II类分子限制性T辅助淋巴细胞和单核细胞。经磁珠分选出的CD4+T细胞仍可保持活性和生物学功能。有研究人员应用免疫磁珠从自然感染的CD4+细胞中分离出了HIV。抗人CD4抗体免疫磁珠还可以用于检测急性T细胞成淋巴细胞白血病,成淋巴细胞淋巴瘤和辅助性T细胞亚群衍生的慢性T细胞淋巴细胞白血病。
Western Blot:Stability: The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37°C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.
Storage: Store at 4°C for frequent use. Stored at -20°C in a manual defrost freezer for one year without detectable loss of activity. Avoid repeated freeze-thaw cycles.
