Background:1. 取100 μl EDTA抗凝的人外周血加入5 ml流式管底;2. 取10 μl标记抗体加入流式管底与全血混匀,室温避光孵育20分钟;3. 加入2ml 1×红细胞裂解液混匀后避光放置10分钟,溶解红细胞(推荐: RBC Lysing Solution[10×]);4. 将样品管置离心机中以1000 rpm离心5分钟后弃上清;5. 加入2 ml PBS洗液重悬细胞,1000 rpm离心5分钟,弃上清;6. 加入0.5 ml PBS洗液重悬细胞,待流式细胞仪检测(样品应在当天上机测定,也可加固定
Molar Function:2D6-B2 单克隆抗体与小鼠和人的CD49f抗原反应,也被称为整合素α6, VLA‐6α 链和 血小板 gpIc. CD49f 是 120 kD 跨膜蛋白. CD49f 与 CD29联系在一起, 与整合素β 1 链, 形成 VLA‐6 复合体; CD49f 与 CD104联系在一起, 与整合素β 4链形成α6β4 复合体. CD49f 在早期T cells, 单核细胞,血小板, 上皮和内皮细胞上表达。 CD49f 也被发现在生发中心B 细胞上表达。2D6-B2 抗体与 人,小鼠,牛细胞的整合素α6反应。这种抗体也被报道在阻断整合素α6与层粘连蛋白结合有功能活性。
Western Blot:Stability: The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37°C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.
Storage: Store at 4°C for frequent use. Stored at -20°C in a manual defrost freezer for one year without detectable loss of activity. Avoid repeated freeze-thaw cycles.
